摘  要:目的:探讨马尾神经受压对腰骶神经元胞体的影响。方法:实验于2002-10/2003-01在西安交通大学第二医院动物试验中心完成。成年健康家犬10只,随机选取2只为对照组,余8只为实验组。对照组打开椎板,置入水囊,不注水。实验组家犬麻醉后打开L7椎板,将硅胶水囊潜行置入L5,6椎板下,注水0.5mL/周,当压力分别达20,60,120mmHg(1mmHg=0.133kPa)和120mmHg延时1周时,随机抽取2只,颈静脉灌注40g/L多聚甲醛处死,切取L6脊髓前角和相应的神经节,用透射电镜观察细胞核染色体的变化,应用缺口末端标记技术观察不同压力时运动神经元细胞凋亡数。结果:整个实验过程中无动物死亡,全部进入结果分析。①透射电镜观察结果:在透射电镜30000倍镜下,对照组及压力≤60mmHg的实验组动物在脊髓前角及神经节均未发现凋亡细胞;当压力≥120mmHg时,脊髓前角细胞核染色体发生边集,染色体发生固缩,且电子密度增强,核形不规则,核膜表面凹凸不平,细胞体积变小,细胞浆浓缩,其内细胞器保存较好,细胞膜保存完整,细胞膜表面微绒毛减少或消失。神经节细胞正常,未发现凋亡细胞。采用原位末端标记法染色标记的凋亡细胞核呈棕褐色着染,细胞核形态呈碎点状,不规则,大小不一致,而非凋亡细胞和阴性对照片细胞核被苏木精复染呈蓝色,核相对较大,形态大小较为一致。②原位末端标记法染色标记的凋亡运动神经元细胞数:压力在20,60mmHg时与对照组无差别(0,0,1±0.23),当压力在120mmHg时脊髓前角有少量凋亡细胞(1±0.12),120mmHg延时1周时脊髓前角运动神经元细胞出现明显凋亡(8±0.67)。结论:成年犬严重压迫马尾神经后,可引起相应运动神经元胞体的死亡,其形式是凋亡。AIM:To study the effect of the cauda equina compression on the lumbosacral neuron soma. METHODS:The experiment was finished in the Laboratory of Animal, Second Hospital of Xi'an Jiaotong University from October 2002 to January 2003.Ten healthy canines were randomly divided into 2 canines in control group and 8 in experimental group.The lamina of vertebra of L7 of the 8 canines in experimental groups and 2 canines in control group was removed after anesthesia,and a water sac made of silica gel was laid under the lamina of L5,6, 0.5 mL water was injected into the sac of 8 canines in experimental groups per week after operation.Two canines selected randomly from the experimental group were injected with 40 g/L formaldehydum polymerisatum through jugular vein when the pressure in the sac respectively reached to 20 mm Hg, 60 mm Hg,120 mm Hg(1 mm Hg=0.133 kPa) and oneweek 120 mm Hg.The spinal cord anterior horn of L6 and its ganglions were cut to observe chromosomal changes of nucleus with electron microscope and in situ endlabeling staining was used to observe the number of apoptotic motor neurons in different pressures. RESULTS:All animals were involved in the result analysis.①The result of electron microscope indicated no apoptosis cells were found in spinal cord anterior horn and its ganglions in control group and experimental group with ≤60 mm Hg pressure in 30 000 times mirror.The chromosome of nucleus in spinal cord anterior horn gathered at the edge with pycnosis;electronic density increased;nucleus indicated anomalistic form with uneven surface; cells dwindled with concintration cell plasm,better organell and intact cell membrane;microvillus of cell membrane decreased or disappeared.The cells in ganglions were normal.The apoptosis nucleuses indicated brown with anomalistic form stained with in situ endlabeling staining, but the nonapoptosis nucleuses indicated blue with bigger and regular form.②There was no significant difference in the number of apoptosis cells between the experimental group
