摘  要:研究去大脑皮层血管对大鼠侧脑室室管膜下增殖分化为新的神经元和胶质细胞的作用和机制。以去大鼠脑皮层血管为模型,以Nestin,BrdU,Tuj1,vimentin,bFGF为指标,采用免疫组织化学和图象分析方法,研究去大脑皮层血管对大鼠侧脑室室管膜下增殖分化为新的神经元的作用和机制。去皮层血管后大鼠侧脑室各水平SVZ的Nestin,BrdU,Tuj1,vimentin,bFGF阳性细胞的数目明显增多,形态显著改变。模型组15天、30天侧脑室前角外侧壁SVZ的Nestin,BrdU,Tuj1,vimentin阳性细胞计数均多于正常组,差异性显著(P<0.01),具有统计学意义。模型组15天和30天前角外侧壁SVZ的bFGF阳性细胞的面密度和光密度均高于正常组,有显著性差异(P<0.01),具有统计学意义。去皮层损伤刺激可引起脑内bFGF水平上调,促进SVZ神经干细胞增殖,并分化新的神经元和胶质细胞,可能参与大脑损伤修复。To study on the effects of cortical devascularization on proliferation and differentiation of neural stem cells located on the subependymal ventricular zone of the lateral ventricle in rat. 65 Adult Wistar male rats, body weight range 200~250 g, devided into normal group and model group randomly. Adopting the immunohistochemistry methods we observe the expression of Nestin, BrdU, Tuj-1, vimentin and bFGF positive neurons located on the lateral wall of anterior horn of lateral ventricular in rat after the operation of cortical devascularization. Cortical devascularization can significantly increase the number of Nestin, BrdU, Tuj-1, bFGF positive neurons located on the lateral ventricle in the rats at each level, compared with normal group, the count number of the Nestin, BrdU, Tuj-1 and vimentin positive neurons located on upper wall of anterior horn and lateral wall on other level of the lateral ventricle in 15d and 30d model group is much greater, P<0.01; Compared with normal group, the area density and optical density of bFGF positive neurons on the SVZ of lateral wall at anterior horn level in the lateral ventricle in model rat brain are all greater, P<0.01. The cortical devascularization can up regulate the bFGF level in the rats’brains, promote the neural stem cells to proliferate and differentiate into neurons and make them to play a role in the repairing of injured cerebral.
