摘 要: 取新生大鼠海马组织进行原代体外分散培养,用MTT比色法检测不同浓度维生素C对神经细胞生长的影响,并观察其形态学变化和分化程度。结果表明,2和20nmol/L维生素C组对神经细胞生长的影响与对照组无显著差别(P>0.05);200nmol/L维生素C组能促进神经细胞的存活,与对照组相比差异显著(P<0.01),进一步观察发现,该处理的细胞突起数目、长度、胞体面积均显著高于对照组(P<0.01);2000nmol/L维生素C组可显著减少细胞的存活数,能引起细胞凋亡。提示200nmol/L维生素C对神经细胞生长发育有一定的促进作用。<Abstrcat>The effects of Vitamin C (V_C) with different concentrations on the growth of hippocampal neurons from newborn rat were detected with MTT method by dissociated cell culture,and the morphological changes and the differentiation degrees were observed.Results indicated that the number of survived neurons in 2 nmol/L and 20 nmol/L V_C groups were almost the same as control group (no V_C in medium (P>0.05).The number of survived neurons in 200 nmol/L V_C group was significantly higher than that of control group (P<0.01),and the survived neurons in 2 000 nmol/L V_C group were scanty,which could lead to cell apoptosis.The number and the length of neuronsal processes as well as the area of neurons in 200 nmol/L group were all increased markedly,enhanced significantly as compared with control.These results indicated that the suitable amount of V_C could improve neuron growth in vitro.
