肺癌相关基因LSCC-3的差异性表达及其与临床和病理的关系

摘　　要：目的研究人肺癌相关基因LSCC3表达与肺癌分期,病理类型,预后之间的关系。方法提取50对肺癌组织和癌旁正常肺组织标本的总核糖核酸(RNA),以人类小核糖体RNA(18srRNA)为内参照行半定量RNA反转录聚合酶链式反应(RTPCR),检测LSCC3在肺癌组织和癌旁正常肺组织中的差异表达,并结合50例患者的临床资料,分析该基因的表达程度与肺癌病理类型、pTNM分期、无瘤生存时间,生存率之间的关系。结果LSCC3基因在肺癌组织中的表达低于癌旁正常肺组织(P<0.01);该基因的表达程度与肺癌患者的性别(P>0.05)、病理类型(P>0.05)、肿瘤分化程度(P>0.05)、病理分期(P>0.05)之间无明显相关;肺癌组织中LSCC3基因高表达患者的无瘤生存时间和生存率高于LSCC3基因低表达患者。结论LSCC3可认为是一个肺癌相关基因,并且可能是一个抑癌基因;LSCC3基因高表达患者预后较低表达者明显改善。Objective To investigate the relationship between the expression of LSCC-3,a lung cancer related gene,and the stage,pathologic styles and prognosis of the patients suffered from lung cancer.[WT5”HZ]Methods A total of 50 couples of samples of lung cancer and normal tissues nearby were used.Total RNA was extracted and semi-quantitative RT-PCR was performed to confirm the differential expression of LSCC-3 in the two different samples.The relationship between the expression of LSCC-3 and the clinical characteristics of lung cancer,such as pathologic styles,pTNM stages,tumor free survival and overall survival time,was studied after all the clinical data were reviewed.Results (1)The expression of the gene LSCC-3 was lower in the lung cancer than in the adjacent lung tissues (P< 0.01);(2)The expression of LSCC-3 was not associated with gender (P> 0.05),tumor pathology (P> 0.05),differentiation (P> 0.05) and tumor stage (P> 0.05);(3) The tumor free survival and overall survival time were longer in the patients with a higher expression of LSCC-3 than that in the patients with a lower expression of LSCC-3.Conclusion The gene LSCC-3 is a lung cancer related gene and a higher expression of LSCC-3 may lead to a better prognosis.
