摘　　要：目的比较AD病人和健康老年人外周血中的抗Aβ抗体特性。方法随机选择AD患者45例和健康老年人51例,①用间接ELISA法测定血清中的抗Aβ抗体滴度,②免疫组织化学ABC法观察AD病人和健康老人血清中的抗Aβ抗体与Tg2576鼠脑内老年斑的结合情况;③不同抗体水平的AD病人和健康老人的血清分别与PC12细胞培养,用MTT法检测PC12细胞的存活率。结果①AD病人和健康老人血清中的抗Aβ抗体分别为(119.78±84.50)μg/ml和(200.44±216.78)μg/ml,差异有显著性意义(P<0.05);②免疫组化发现用AD病人血清作一抗,抗Aβ抗体不能很好地显示已经形成的老年斑;③虽然AD病人血清中抗Aβ抗体较高,对PC12细胞的保护作用反而下降,且与Aβ42的剂量无相关性;而健康老人的血清仍能表现出较强的PC12细胞保护作用。结论AD病人血清中抗Aβ抗体滴度下降,与老年斑结合能力和对细胞的保护能力也下降。Objective To investigate the characteristia of anti-Aβ anti bodies in patients with AD. Methods Serum were obtained from 45 AD patients and 51 healthy elderly individuals. The titer of anti-Aβ antibodies in serum was m easured using an indirect ELISA. The senile plaque was detected by immunohistoch emical staining, using the serum as the first antibody. PC12 cells were plated i n 96 wells. Aβ42 and the serum were added to cultures and incubated for 4 days. Cells survival rate was measured by MTT assay. Results The titer of anti-Aβ a ntibodies was significantly lower in serum from AD patients than the healthy eld erly individuals (P<0.05). The serum from AD patients did not recognize the seni le plaques in the brain section from Tg2576 mouse. Furthermore, the lower anti-A β antibodies in the serum from AD patients and healthy elderly individuals can protect the PC12 cells. However, the higher anti-Aβ antibodies in AD patients c an not inhibit the cytotoxicity of Aβ42. Conclusion AD patients may have speci fic immune impairment for the production and property of anti-Aβ antibodies.
