摘　　要：目的研究Tenascin(TN)在原发性肝细胞癌(HCC)中的表达,探讨TN与HCC浸润和转移的关系。方法分别采用免疫组织化学SP法和逆转录聚合酶链反应(RTPCR)方法检测42例HCC、10例肝硬化和7例正常肝组织内TN的表达情况,并结合临床病理指标进行分析。结果TN在HCC、肝硬化和正常肝脏组织中的阳性例数分别为26、2和0例,其差异具有显著性意义(P<0.05);HCC组织内TNmRNA的表达强度明显高于肝硬化组织(t=2.793,P<0.05);TN在有包膜侵犯组、病理分级Ⅲ～Ⅳ组及有转移HCC中的阳性表达率及强度明显高于无包膜侵犯组(P<0.05)、病理分级Ⅰ～Ⅱ级组(P<0.05)以及无转移组(P<0.01)。结论TN在HCC组织中的表达上调与HCC的浸润和转移有关,检测其表达有助于判断肝癌的恶性程度和转移性。Objective To study the expression of Tenascin in human hepatocellular carcinoma (HCC) and its relationship between invasion and metastasis of HCC.Methods By using the method of immohistochemical staining (SP) and reverse transcription-polymerase chain reaction (RT-PCR), specimens from 42 cases of HCC, 10 cases of hepatic cirrhosis (HC) and 7 normal liver tissues (NL) were studied. The relationship between the Tenascin expression and HCC pathological features was evaluated.Results Tenascin positive cases in the HCC, HC and NL tissues was 26, 2 and 0 respectively, with the significant difference between HCC and HC as well as NL (P<0.05). The expression of Tenascin mRNA in HCC tissues was much higher than that in HC (t=2.793, P<0.05). Overexpression of Tenascin in the HCC was related to the invasion of tumor envelop (P<0.05), Edmondson grades (P<0.05) and tumor metastasis (P<0.01).Conclusion Tenascin may play an important role in invasion and metastasis of HCC. It can be regarded as an indicator for malignancy and metastasis of HCC.
