摘 要:目的研究环氧化酶1、2(Cox1、Cox2)在缺血性视网膜病变中的作用。方法高浓度氧诱导C57BL/6J小鼠建立视网膜缺血性病变动物模型。苏木精伊红染色,观察高氧组和正常组视网膜切片新生血管形成,逆转录聚合酶链式反应(RTPCR)方法半定量测定高氧组和正常组Cox2及Cox1mRNA的表达。结果正常组每只眼视网膜新生血管内皮细胞核的平均数为(0.36±0.67),高氧组每只眼均可见突出内界膜的新生血管内皮细胞,发生率为100%,每只眼视网膜新生血管内皮细胞核的平均数为(251.23±22.25)。高氧组Cox2表达为正常组的4.6倍(P<0.05),两组Cox1mRNA表达无明显差异(P>0.05)。结论Cox2表达水平的上调可能与视网膜新生血管的形成有关。Objective To detect the expression of cyclooxygenase-2 (Cox-2) mRNA and Cox-1 mRNA in ischemic retinopathy.Methods Mouse models of hyperoxia-induced ischemic retinopathy were established in C57BL/6J mice. The mice were divided into control group and hyperoxic group. The nuclei of new vessel buds extending from the retina into the vitreous body in different groups were counted and compared under the light microscopy. Fibrovascular tissues were obtained from 17 day-old mice in different groups. Reverse transcripition polymerase chain reaction (RT-PCR) analysis were performed to detect the expression of Cox-2 mRNA and Cox-1 mRNA.Results The average number of nuclei of new vessel buds in each eye on control and hyperoxic groups were (0.36±0.67) and (251.23±22.25) respectively. In hyperoxic group, neovascular endothelial cells could be seen in each eye. The mRNA experssion levels of Cox-2 were increased by 4.6-fold as compared with control group, while there was no significant difference in the Cox-1 mRNA expression between the two groups.Conclusion The up-regulation of the Cox-2 mRNA expression might contribute to the neovascularization in ischemic retinopathy.
