摘  要:目的观察壳聚糖磷脂复合物对痴呆模型大鼠的治疗效果,初步探讨其作用机制。方法将50只SD大鼠随机分为5组(每组10只),分别为:空白对照组(1组)、假手术组(2组)、痴呆模型组(3组)、脑复康治疗组(4组)和壳聚糖磷脂复合物治疗组(5组)。分别观察各组大鼠Morris水迷宫逃逸潜伏期、脑电超慢涨落图S谱线、投射电镜下脑组织的超微结构及免疫组织化学及图像分析检测脑组织乙酰胆碱酯酶和基底神经生长因子含量。所得资料应用SAS6.12进行统计学处理。结果壳聚糖磷脂复合物可以缩短痴呆鼠逃逸时间,提高S5谱线频率,与痴呆模型组(P<0.01)和脑复康治疗组(P<0.05)相比均有统计学意义;可以恢复提高大脑皮层和基底核胆碱能神经元的数量、面积和光密度(P<0.01),减少损伤脑组织变性水肿;脑复康治疗组和壳聚糖磷脂复合物治疗组脑内神经生长因子含量较痴呆模型组有所提高,但无统计学差异(P>0.05)。结论壳聚糖磷脂复合物能改善痴呆大鼠的学习和记忆功能,可能与阻止和修复胆碱能神经元损伤,增加乙酰胆碱含量,以及参与脑组织抗炎免疫反应有关。Objective To study the effect and the mechanism of Chitosan-phsospholipid complex compared piracetam on Alzheimer's disease (AD) models in rats.Methods Fifty rats were divided into five groups randomizedly and named the control rat group (the 1st group),the sham rat group (the 2nd group),the AD model rat group (the 3rd group),the piracetam therapy rat group(the 4th group) and the Chitosan-phsospholipid complex therapy rat group(the 5th group).Every group has 10 rats. The escape latency of morris water maze test,the frequency of S_5spectral line in encephalofluctuogram technology,the quantity and area of AchE and NGF in immune histochemistry staining,the assaying cellular and morphologi morphologic changes of nucleus basalis of Meynert were analyzed analysis by SAS in each group.Results Chitosan-phsospholipid complex could shorten the escape latency and improve the frequency of S_5 spectral line in AD model rats group compared with the AD model rat group(P<0.01) and the piracetam therapy rat group(P<0.05).immume histochemistry sraining and assaying showed AchE markedly decreased in the model rat group,and the piracetam therapy rat group and the 5th group had significantly increased compared with the 3rd group,there were few pathologic abnormality existed in therapy rat group compared with AD model rat group.There were no statistic changes of NGF between the 3rd and the 4th and the 5th group.Conclusion The Chitosan-phsospholipid complex,improving the AchE contents of the neuron and promoting injured nerve cell recover,has an evidence therapy-effect on AD model rats.
