摘 要:目的:研究能检测食品中罂粟碱含量的酶免测试盒。方法:利用研制的罂粟碱抗体,罂粟碱酶标抗原以及相关的ELISA其它试剂,进行测试盒的各项技术指标的确证。结果:结果表明:测试盒的最低检测限:0.1ng/ml;定量检测范围:0.1~20ng/ml;精密度(批内CV%):5.7%~9.1%;样品加标回收率79%~123%;与类似生物碱的交叉反应率很低(<1.7%);测试盒4℃下保存期可达6个月以上;与HPLC法和免疫胶体金法对比实验表明,有良好的一致性。结论:研制的罂粟碱酶免测试盒能定性定量检测食品中的罂粟碱含量。Objective:To develop an ELISA kit for detecting papaverine in food.Methods:The method-indices of the kit was validated by using of the preparing papaverine antibody and the papaverine-HRP enzyme conjugate and other relevant reagents of ELISA.Results:The detection limit for papaverine of the kit was 0.1 ng/ml,quantitative range of the kit was 0.1~20 ng/ml, coefficients of variation (CV%) was 5.7%~9.1%, the rate of recoveries was 79%~123%, the cross-reactivities relative to homologous alkaloid were low (<1.7%), validity of the kit under 4℃ was over 6 months.Comparision with HPLC and colloid gold immunoassay showed a better consistent result.Conclusion:The ELSIA kit for papaverine can quantitate or semi-quantitate the content of papaverine in food.
