摘  要:目的研究不同间隔时间多次化学性缺氧对大鼠海马CA1区神经元凋亡及Bcl2蛋白表达的影响。方法应用原位末端标记技术及免疫组化技术,观察分析对照组(A组)、3硝基丙酸(3NPA)连续给药组(B组)、3NPA间隔给药组(C组)大鼠海马CA1区神经元凋亡及Bcl2蛋白表达情况。结果(1)大鼠海马CA1区神经元凋亡B组较A、C组有显著增加(均P<0.05),而A组与C组比较差异无显著性(P>0.05);(2)Bcl2蛋白免疫反应强度C组较A、B组增强非常显著(均P<0.001),而A组与B组比较差异无显著性(P>0.05)。结论连续多次应用小剂量3NPA可使大鼠海马CA1区神经元凋亡明显增加,间隔多次应用小剂量3NPA对神经元有保护作用。Objective To investigate the effects of multiple chemical anoxia caused by low dose of 3-nitropropionic acid (3-NPA) with different intervals on apoptosis and expression of Bcl-2 protein in hippocampal CA1 region of rats.Methods Using in situ end labeling method and immunohistochemical analysis, the authors observed and analysed the number of apoptotic cells and immunoreactivity of Bcl-2 protein in hippocampal CA1 region of the rats, including control group (group A), short interval group (group B), long interval group (group C).Results (1) The apoptotic cells of group B were significantly more than those of group A and C(all P<0.05), but there was no difference between group C and group A (P>0.05). (2) Bcl-2 immunoreactivity was much higher in group C than in group A and B (all P<0.001), and no remarkable difference was found between group B and group A (P>0.05).Conclusions Consecutive use of low dose of 3-NPA may increase neuronal apoptosis in CA1 region of rats, but interval use of low dose of 3-NPA may play neural protective effect.
