人鼻咽癌细胞胞质内骨架系统的观察和维胺酸作用后对其影响的研究

摘　　要：用原位包埋法、细胞质骨架生物化学抽提、免疫组织化学和组织化学等方法，对正常人胚肺成纤维细胞、非洲绿恒河猴肾上皮细胞和人鼻咽癌细胞胞质骨架的形态学特征作了比较观察，并就维胺酸对人鼻咽癌细胞胞质骨架的形态分布、角蛋白、波形纤维蛋白等骨架成分的影响进行了研究。结果表明，与正常细胞相比，人鼻咽癌细胞胞质内骨架系统发育不良。维胺酸作用后，癌细胞中细丝状骨架纤维增多，并出现应力纤维束样排列，角蛋白增加。The cytoskeleton system of normal and malignant cells was observed by using of routine investment in situ,biochemical extraction,immunohistochemistry and histochemistry in this experiment.The effects of RII( 4-hydroxy-carbophenyl retinamide )on the distribution of cytoskeleton system,and the expression of keratin and vimentin in human nasopharyngeal carcinoma cells (HNPC)were studied.The results demonstrated that the cytoskeleton system of HNPC cells is maldeveloped as compared with that of fibroblast cells and kidney epithelial cells. The amount of cytoskeletal fibers increased and stress fiber appeared in HNPC cells after RII treatment.This study suggests that RII can improve expression of keratin in HNPC cells.
