摘 要:目的探讨层粘连蛋白(Laminin,LN)在大鼠脑缺血再灌注中的作用。方法制备大鼠脑缺血再灌注(MCAO/R)模型,分为假手术组与缺血再灌注组;缺血2h后依照再灌注时间点的不同分为2h、6h、12h、24h、3d及7d共6组。各时相点取材,免疫组化观察脑组织LN的表达变化,并使用多媒体彩色病理图像分析系统进行定量分析。结果LN的表达于再灌注2h开始降低,24h时达最低,第3日时开始回升。结论LN可能参与了缺血再灌注血管源性脑水肿的形成与发展,并且可能参与了再灌注损伤后新生血管的生成、血管重塑及侧支循环的建立。Objective To study the function of Laminin in brain ischemia/reperfusion in rats. Methods Rat model of ischemial/reperfusion of cerebral middle artery was established. Rats were randomly divided into sham-operation group and ischemia/reperfusion group. The ischemia/reperfusion group was further divided into six subgroups according to reperfusion time interval such as 2h?6h?12h?24h?3d and 7d. Immunohistochemical staining method and color image analysis were used and the expression of Laminin in the ischemic region were measured. Results The expression of Laminin was decreases in MCAO2h group and reaches the minimum in MCAO2h/R24 group. It started to increase 3 days later. Conclusions Laminin probably has close relationship with the formation and progression of vascular brain edema. Laminin can promote blood vessel proliferation surrounding ischemic zone and help damage recovery.
