摘  要:目的 研究神经突触前膜胞内蛋白(Munc18)抗体致痫大鼠的机制。方法60只SD大鼠分为4组,在实验组大鼠的海马CA1区间断注射Munc18抗体,共9次。监测大鼠的脑电图和痫样行为。10周后全部大鼠处死取脑,切片做HE、尼氏和TUNEL染色后观察。结果 实验组12只大鼠中,10只出现异常脑电,5只在6周之后仍然存在;9只出现1-4级不等的痫样行为,6周后仍有4只存在痫样行为;大鼠脑片显示海马区神经细胞数量明显减少(155±20,P<0.01),而凋亡细胞数量明显增多(16.80±3.32,P<0.01),伴有胶质细胞增生及细胞形态异常。结论 Munc18抗体能慢性点燃致痫大鼠,其机制可能与其诱导整个海马区神经细胞的凋亡和丢失有关。Objective To investigate the mechanism of presynaptic protein (Munc18) antibody in inducing epilepsy on rat. Methods Munc18 antibody was continuously micro-injected into CA1 area of hippocampus in Sprague-Dawley (SD) rat (the injection times were nine). The potential epileptic seizure was evaluated according to both electroencephalogram (EEG) and behaviors. HE, Nissle and TUNEL staining were applied on rat brain sections as to detecting the cell damage and apoptosis after tenth week. Results Ten of twelve rats produced epileptic EEG, five cases among these ten rats maintained abnormal EEG after the sixth week since injections. Nine of the above twelve rats had epileptic seizure according to Racine grade one to four, four cases among these nine rats kept seizure after the sixth week since injections. The number of neurons decreased (155±20,P<0.01) and TUNEL-positive neurons increased (16.80±3.32,P<0.01) in whole hippocampus area. Neurons appeared to be morphologic abnormality and glial cells proliferated. Conclusions Munc18 antibody might induce epilepsy by chronic kindling way on rat,suggesting that the mechanism might be linked to neurons’s apoptosis and lose in hippocampus area.
