摘 要:目的:观察氯通道阻断药尼氟酸(NFA)对大鼠体外心脏缺血再灌注性心律失常及心功能的影响。方法:采用大鼠Langendorff体外心脏灌注模型,阻断冠状动脉,模拟心肌缺血,观察心功能指标、心率和心律失常积分值的变化。结果:NFA(10μmol/L)可使缺血性心律失常分数值明显降低(P<0.01),但对再灌注性心律失常无明显影响;NFA对体外大鼠心脏具有明显的负性变时和变力作用(P<0.05),且呈剂量依赖性。结论:氯通道阻断药NFA具有明显的抗缺血性心律失常作用。Objective: To investigate the effects of niflumic acid (NFA), a blocker of chloride channels, on ischemia-reperfusion arrhythmia and heart function in rats. Methods: The study was carried out in Langendorff perfused rat hearts subjected to coronary artery occlusion in which the severity of arrhythmias, index of heart function and heart rate were measured. Results: The severity of ischemia induced arrhythmia (arrhythmia score) was lower significantly in NFA(10mmol/L ) pertreated hearts than the control (P<0.01), but NFA had no effects on reperfusion-induced arrhythmia. In addition, NFA inhibited significantly the cardiac contraction and heart rates in a dose-dependent manner (P<0.05). Conclusion: The significant antiarrhythmic role of NFA, as a chloride channel blocker, was confirmed.
