GNB3基因C825T多态同哈萨克族人群肥胖的关联分析

摘　　要：目的探讨鸟苷酸结合蛋白基因β3亚单位(G-proteinβ3Subunit,GNB3)第10外显子C825T多态同中国哈萨克族人群肥胖的关系。方法应用PCR技术及限制性长度多态(RFLP)技术在277例哈萨克族正常体重者,121例超重者及102例肥胖者中检测GNB3基因C825T多态性。结果GNB3基因C825T多态3种基因型分别为TT、CT及CC。其中CT、TT基因型分布频率在正常体重组、超重组及肥胖组分别为46.9%,54.5%,54.9%,23.4%,16.6%,20.6%,基因型分布在组间无显著性差异(P=0.39)。T等位基因分布在正常体重组、超重组及肥胖组分别为46.9%,43.8%,48.0%,组间也无显著性差异(P=0.63)。将正常体重及超重者按高血压及正常血压分层,发现超重及肥胖的高血压者TT基因型频率(26.2%)较正常体重的高血压者(14.7%)有增高趋势(P=0.061)。结论GNB3基因825T等位基因可能不是中国哈萨克族肥胖的主要遗传易感因子,但TT基因型可能在超重的高血压者中有一定作用,此基因在中国其他人群肥胖合并高血压中的作用有必要进行深入研究。Purpose Present study was to investigate the relationship between C825T polymorphism in the G-proteinβ?3 Subunit (GNB3) gene and obesity in Kazakh. Methods The variants C825T of GNB3 gene were determined using PCR and RFLP technique in 277 Kazakh normal weight,121 overweight,and 102 obesity subjects. Results The distributions of CT and TT genotypes were 46.9%,54.5%,54.9%,23.4%,16.6%, 20.6% in normal weight,overweight,and obesity subjects.The T allele was 46.9% in normal weight subjects,43.8% in overweight subjects,and 48.0% in obesity subjects.There were no signi-ficant differences of both genotype distribution(P=0.39) and allele frequencies(P=0.63) among normal weight subjects,overweight subjects and obesity subjects.When normal weight subjects and subjects with BMI≥25.0 kg/m2 were stratified with blood pressure,an increased trend of TT allele can be observed in overweight hypertensive subjects(26.2% vs 14.7%),despite statistically increased rate was not observed (P=0.061). Conclusions These results suggested that the 825T allele of the GNB3 gene does not confer a major increased risk for obesity in the Kazakh populations,but TT genotype may play some roles in overweight hypertensive subjects and further study needs to be conducted.
