摘 要:目的研究凝胶化改性壳聚糖的抑菌及抗感染作用。方法将壳聚糖行凝胶化改性,制成壳聚糖凝胶和壳聚糖膜。采用琼脂打孔扩散法,应用1%、2%、4%的壳聚糖凝胶进行体外抑菌试验。抗感染试验在大鼠蚓突盲端刺孔制作腹腔感染模型,再将大鼠随机分成实验组和对照组,实验组应用壳聚糖膜覆盖带漏口的蚓突盲端,对照组不用膜,术后2周和4周观察腹腔感染的严重程度。结果抑菌试验三种不同浓度的壳聚糖对所用的五种细菌均有明显的抑制作用,但其对金黄色葡萄球菌、大肠杆菌、枯草杆菌和绿脓杆菌的抑菌环直径均显著小于哌拉西林和林可霉素(P<0.05);哌拉西林和林可霉素对白色念珠菌均无效,但三种浓度的壳聚糖组对白色念珠菌均形成明显的抑菌环。抗感染试验术后2周和4周,实验组的腹腔感染程度均显著轻于对照组(P<0.05)。结论凝胶化改性壳聚糖不但对一般性感染菌具明显的抑制作用,对腹腔感染具一定的预防作用,且能抑制白色念珠菌的生长,具潜在的医用开发价值。Objective To study the effects of gelatinizedly-reshaped chitosan on the cultured bacteria in vitro and infection of abdominal cavity in vivo. Methods Chitosan reshapedin gelatinization, formed into gelatum and film. Its in vitro antibacterial ect was measured by the method of holing agar medium and diffusing drug in the holes. The concentrations of applied chitosan gel were 1%, 2% and 4%. To study the anti-infection effect of the chitosan film, 40 Sprague-Dawley rats were randomly divided into two groups, experimental group and control group. All the rats were pricked holes in the caecums of vermiform processes. The caecums of experimental group were covered with chitosan film, and in the control group, with no film. Two and four weeks after the treatment, the infectious severities in the abdominal cavities of rats were determined. Results All of the chitosan gel of three different concentrations demonstrated remarkable antibacterial effects. To the Staphylococcus aureus. Escherichia coli, Bacillus hay and Bacillus pyocyaneus, Antibacterial rings of the three chitosan groups were significantly smaller than the positive groups(with piperacillin and lincomycin, P < 0.05). To the Candida albicans, piperacillin and lincomycin manifestated no antibacterial effect and in the three chitosan groups, the effect was significant, in the anti-infection experiments of rats, the infectious severity of experimental group decreased significantly when compared with the control group both two and four weeks after the treatment(P < 0.05). Conclusion Gelatinizedly-reshaped chitosan possesses antibacterial effect not only on the general bacteria in vitro and abdominal infection in vivo, but also on the Candida a/bicans. It has potential great values for development in the biomedical area.
