摘  要:目的研究hB7-1基因转染对人卵巢癌SKOV3细胞生物学特性及MHCIb和HLA-DR抗原表达的影响。方法以腺病毒为载体将hB7-1基因转染SKOV3细胞,噻唑蓝还原法检测SKOV3细胞生长曲线;流式细胞技术观察转染前后细胞周期改变以及hB7-1、MHCIb、HLA-DR分子表达;电镜观察细胞形态学变化。结果hB7-1基因转染人卵巢癌SKOV3细胞前后细胞生长曲线无明显差异(P>0.05);SKOV3细胞感染病毒48h后在相同条件下形态上较感染前无明显改变;hB7-1基因转染48h后,SKOV3细胞表面MHCIb、HLA-DR抗原表达较转染前分别增高1.10±0.22和1.03±0.17,80%细胞表面有较高水平hB7-1基因表达,差异显著(P<0.05)。结论hB7-1基因转染人卵巢癌SKOV3细胞后对细胞生长能力、细胞周期、细胞超微结构变化等生物学特性无明显影响,可显著增强人卵巢癌SKOV3细胞表面的MHCIb和HLA-DR抗原表达,为实现卵巢癌基因治疗提供理论基础。Objective To investigate the influence of hB7-1 transfection on the SKOV3 biological characteristics of human ovarian carcinoma and the expression of antigen MHC Ib and HLA-DR.Methods hB7-1 was transfected to SKOV3 with adenovirus as vector,the cellular growth curve was detected with MTT,the cellular cycle and the expression of hB7-1 and HLA-DR were observed with flow cytometry (FCM),and the cellular morphologic changes were observed with electrical microscopy.Results The cellular growth curve had no distinctive signifcance before and after transfecting hB7-1 into SKOV3 (P>0.05),and the morphology changes of SKOV3 were not significant before and 48 h after hB7-1 transfecting in the same condition.48 h after hB7-1 transfecting the expression of MHC Ib and HLA-DR increased by 1.10±0.22 and 1.03±0.17,respectively.80% cells expressed hB7-1,with significant difference (P<0.05).Conclusion hB7-1 transfected to SKOV3 does not change the cellular growth ability,cellular cycle and cellular ultrastructure,which may obviously promote the expression of MHC Ib and HLA-DR,providing theoretical basis for the gene therapy of ovarian carcinoma.
