摘  要:目的分析与N-糖链合成相关β-1,4半乳糖基转移酶-Ⅰ、Ⅱ、Ⅴ(β1,4-galactosyltransferaseⅠ、Ⅱ、Ⅴ,β-1,4-GalT-Ⅰ、Ⅱ、Ⅴ)mRNA在正常脑组织和各级人星形胶质细胞瘤的表达变化。方法采用RT-PCR方法,分析β-1,4-GalT-Ⅰ、Ⅱ、Ⅴ在人星形胶质细胞瘤的表达存在。以扩增的3种同工酶cDNA片段作为探针进行标记,Northernblot分析其mRNA在正常脑组织和各级人星形胶质细胞瘤中的表达变化。结果RT-PCR检测到β-1,4-GalT-Ⅰ、Ⅱ、Ⅴ在人星形胶质细胞瘤中的表达存在。northernblot发现β-1,4-GalT-Ⅱ、Ⅴ在正常脑组织中有表达,但β-1,4-GalT-Ⅱ的表达较低,而β-1,4-GalT-Ⅰ在正常脑组织中表达极低。β-1,4-GalT-Ⅰ、Ⅱ、Ⅴ在星形胶质细胞瘤中的表达显著高于正常脑组织。其中,β-1,4-GalT-Ⅰ、Ⅱ在各级胶质瘤中的表达没有明显差异。而β-1,4-GalT-Ⅴ的表达随胶质瘤分级的增高而增加。结论N-糖链合成相关的β-1,4-GalT-Ⅰ、Ⅱ、Ⅴ在人脑星形胶质细胞瘤发生过程中的表达不同,尤其是β-1,4-GalT-Ⅴ的表达与人星型胶质瘤的恶性程度显著相关。Objective To analyze the changes of mRNA expression of β1,4-galactosyltransferase Ⅰ,Ⅱ and V(β-1,4-GalT-Ⅰ,Ⅱ and V) associated with synthesis of N-linked oligosaccharides in normal brain and human astrocytomas.Methods The fragments of β-1,4-GalT-Ⅰ,Ⅱ and V were obtained by RT-PCR from total RNA of human astrocytomas.The amplified cDNA fragments were used as the probes to analyze the expression changes of β-1,4-GalT-Ⅰ,Ⅱ and V in normal brain and human astrocytomas by Northern blot.Results The RT-PCR results showed that β-1,4-GalT-ⅠmRNA was very rare in normal human adult brain but was detectable in grade Ⅰ,Ⅱ,Ⅲ and IV of astrocytomas.The level of β-1,4-GalT-Ⅱ mRNA was increased in grade Ⅱ,Ⅲ and IV of astrocytomas while only a trace amount was found in normal brain.β-1,4-GalT-V mRNA existed in normal brain and exhibited an increased expression with the process of astrocytomas differentiation.The highest level was found in grade IV of astrocytomas.The expressions of three kinds of β-1,4-GalT mRNA were differently changed in human astrocytomas respectively.Conclusions The results indicate that the increase of galactosylation in N-glycans of astrocytomas may be caused by the alterations of the expression of β-1,4-GalT-Ⅰ,Ⅱ and V, and the malignant degree of astrocytoma is significantly correlated with the expression of β-1,4-GalT V.
