摘 要:目的建立大鼠大肠癌动物模型,观察肌醇六磷酸(植酸)对诱癌大鼠大肠组织抗氧化活性的影响。方法30只4周龄雄性Wistar大鼠按体重随机分为植酸组和对照组,每组15只。植酸组饮水添加2%植酸钠,对照组自由饮水。2组大鼠均给予皮下注射1,2-二甲肼(DMH),观察肿瘤发生率、肿瘤的数量及体积;测定大肠组织的超氧化物岐化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性和丙二醛(MDA)含量。结果植酸组大肠癌发生率与对照组相比差异无显著性,植酸组平均每只鼠的肿瘤个数、肿瘤体积均显著低于对照组(P<0.01,P<0.05);植酸组大鼠大肠组织的SOD、GSH-Px活性比对照组显著升高(P<0.01),MDA含量显著降低(P<0.01)。结论植酸可降低诱癌大鼠大肠肿瘤发生的危险,可能通过其抗氧化作用发挥抗肿瘤作用。Objective To evaluate the effect of inositol hexaphosphate(InsP6)on 1,2-dimethylhydrazine(DMH)- induced large bowel antioxidative activity in Wistar rats and discuss the mechanism underlying the anti-neoplastic potential of InsP6. Methods Thirty four-week-old male Wistar rats were randomly divided into two groups, with 15 rats in each group. The treatment group were given 2% Na-InsP6 supple- mented water, and the control group received tap water. The rats were given a weekly injection of DMH to induce colon carcinoma. The number of total macroscopically visible neoplasms of large bowel was counted and the volume of individual neoplasmas was calculated. Superoxide dismutase(SOD), glutathione peroxidase(GSH-Px)and maleicdialdehyde(MDA)of large bowel were analyzed. Results The incidence of tumors in Wistar rats treated with DMH plus InsP6 was not significantly different with rats treated with DMH without InsP6. However, InsP6 significantly decreased the number of tumor and tumor volume(P < 0.01, P < 0.05, respectively). Treatment of rats with Insp6 significantly raised SOD and GSH-Px activities(P < 0.01)and decreased MDA content(P < 0.01). Conclusion Na-InsP6 can effectively reduce the risk of large intestinal cancer in Wistar rats. InsP6 may exert an antineoplastic effect with its antioxidant function.
