摘 要:在HL-60白血病细胞的体外培养中,组胺H_2受体激动剂4-甲基组胺(10 ̄(-8)-10 ̄(-4)mol/L)对HL-60细胞的增殖具有轻微的抑制作用,而组胺H_2受体拮抗剂雷尼替丁(10 ̄(-8)-10 ̄(-4)mol/L)对HL-60细胞的增殖具有较强的抑制作用。但用10 ̄(-6)mol/L的雷尼替丁预处理HL-60细胞则可部分地拮抗4-甲基组胺(10 ̄(-8)-10 ̄(-4)mol/L)对HL-60白血病细胞的抑制作用。The results obtained from liquid suspension culture,semisolid culture and  ̄3H-TdR incorpo-ration into HL-60 leukemia cells indicated that histamine H_2 receptor agonist 4-methylhistamine(4-MH) had slight suppressive effect on HL-60 leukemia cells when the 4-MHdoses were 10 ̄(-8)-10 ̄(-4)mol / L.The IC_50 of 4-MH in HL-60 colony culture was 1.74 ×10 ̄(-5)mol / L. Histamine H_2 receptor antagonist ranitidine exerted its suppressive effect onHL-60 leukemia cells when ranitidine 10 ̄(-8)mol / L to 10 ̄(-4)mol / L were added.The IC_50 ofranitidine in HL-60 colony culture was 4.24×10 ̄(-8)mol / L.Therefore, histamine H_2 receptor an-tagonist ranitidine exerted greater suppressive effect on HL-60 leukemia cells than histamine H_2receptor agonist 4-MH did.When HL-60 leukemia cells were pretreated with ranitidine10 ̄(-6)mol / L for 0.5h,which had no supressive effect on HL-60 cells,the inhibitory effect of4-MH(10 ̄(-8)-10 ̄(-4)mol / L) on HL-60 leukemia cells could be antagonized to some extent.Thesephenomena suggest that histamine H_2 receptor agonist 4-MH and its antagonist ranitidine mayexert their suppressive effects on HL-60 leukemia cells through different mechanisms whichshould be clarified in further experiments.
