摘  要:目的:研究钙通道拮抗剂尼莫地平与血管内皮生长因子(VEGF)及血小板洐生生长因子(PDGF)在视网膜新生血管形成中的相互作用。方法:将SD幼鼠随机分为5组:正常对照组,模型组,球后注射药物大剂量组、中剂量组及小剂量组。建立高浓度氧气诱导的SD幼鼠增殖性视网膜病变模型(OIR),后3组分别双眼球后注射尼莫地平(0.2mg/ml)各10、5、2μl,隔日1次×3次。取实验幼鼠眼球作普通病理切片及免疫组织化学检测,分别检测视网膜新生血管芽内皮细胞数目及VEGF与PDGF的表达。结果:模型组视网膜新生血管芽内皮细胞数目及VEGF和PDGF的表达较正常对照组明显增加(P<0.01),双眼球后注药大、中剂量组较模型组均明显减少(P<0.01),小剂量组则无明显变化。结论:VEGF和PDGF能够促进视网膜新生血管形成,一定剂量的钙通道拮抗剂尼莫地平则能抑制视网膜新生血管的形成。尼莫地平在某种程度上能抑制VEGF及PDGF的表达。Objective: To study the interaction between the calcium channel antagonist nimodipine and growth factors (vascular endothelial growth factor [VEGF] and platelet-derived growth factor[PDGF]) during the formation of new retinal vessels. Methods: The hyperoxia model was induced by proliferative retinopathy (OIR) in newborn Sprague-Dawley (SD) rats. SD rats (2 d after birth) were randomized into 5 groups: normal control group, pure OIR group and group 3, 4 and 5, where the animals received retrobulbar injection of nimodipine 10 μl, 5 μl and 2 μl once every 2 d for 3 times, respectively. Both eyeballs of newborn rats were made into common pathological sections and detected by immunohistochemistry method to count the nuclei of proliferative retinal vessel cells and to investigate the expression of VEGF and PDGF in retina. Results: The nuclei of proliferative retinal vessel cells and the expressions of VEGF and PDGF in pure OIR group increased significantly compared to those of normal control group(P<0.01). The above parameters decreased significantly in group 3 and 4 as compared with those in pure OIR group (P<0.01), and there was no significant decrease in group 5. Conclusion: VEGF and PDGF can induce the formation of new retinal vessels, and calcium channel antagonist nimodipine can inhibit the formation. Nimodipine can inhibit the expression of VEGF and PDGF to a certain extent.
