摘  要:目的观察PB19对离体大鼠心室肌细胞的动作电位(AP)的影响,研究PB19对大鼠冠状动脉缺血再灌注损伤的作用。方法制备大鼠离体心肌模型,记录并观察PB19对离体大鼠正常心肌动作电位的影响;制备在体大鼠心肌缺血及再灌注模型,观察PB19对大鼠缺血性心律失常的保护作用。结果PB19明显延长离体大鼠心室肌细胞复极50%及90%时的动作电位时程(APD50、APD90,P<0.01);大鼠冠脉结扎前后分别给予PB19均缩短VTD(室速持续时间,P<0.05),降低室速(VT)、室颤(VF)发生率和死亡率(P<0.01),并呈剂量依赖性。结论PB19未对Vmax产生影响,但延长了APD。PB19降低了大鼠冠状动脉结扎诱发的室性心律失常和死亡率。这种作用可能与PB19延长动作电位时程有关。Aim To investigate the effects of PB-19 on action potential (AP) of ventricular cells in vitro and ischemia and reperfusion induced arrhythmias in vivo in rats. Methods ① We established the model of isolated rat right ventricular wall and perfused the models with normal Tyrode’s solution and PB-19.Microelectrode was inserted into the cells to record the AP. ② We ligated the left anterior descending coronary artery of rats to study the effects of PB-19 on ischemia and reperfusion induced arrhythmias. 50 rats were divided into 5 groups randomly and were given iv. injection with normal saline and PB-19 of different concentrations respectively. We recorded Ⅱ ECG of the rats. Results ①In vitro models, the APD_ 50、APD_ 90 of PB-19 group were significantly longer than that of control group respectively (P<0.01).②The VTD,VT rate, VF rate and mortality of PB-19 groups were significantly lower than that of control group respectively . Conclusion PB-19 has protective effect on ischemia and reperfusion induced arrhythmias of rats.
