摘　　要：目的用Her2/neu蛋白基因重组腺相关病毒(rAAV-Her-2/neu)转染人外周血中树突状细胞(dendriticcell,DC),并鉴定其表面分子的表达,及同种混合淋巴细胞反应。方法采用Ficol分离人外周血中的单个核细胞,在6孔板上培养。细胞分2组,分别加入病毒液和293细胞冻溶液。流式细胞仪检测DC表面标志。应用MTT法检测同种混合淋巴细胞反应。结果两组细胞均高表达CD80,CD83,CD86,HLA-DR,较低表达CDIa,CD40,两组无明显差异(P>0.05)。结论rAAV-Her-2/neu转染DC刺激T细胞增殖能力明显高于非转染组DC(P>0.05)。Objectives To tiansinfect Her2/neu gene into dendritic cells (DC) in human peripheral blood by recombination adeno - assoi ciated vims (rAAV) vector, and to check the surface markers of DC and the mixed lymphocyte reaction. Metheds The mononuclear cells in healthy person peripheral blood were isolated by Ficol- Hypaque density gradient separation.The DC were divided into two groups.The rAAV were added to one group and 293 cells added to the other group.The surface markers of DC were determined by flow cytometry and the mixed lymphocyte reaction was determined by MTT. Results There were high expressions of CD 80, CD 83, CD 86 and HLA - DR, and Poor expressiond of CD1α, CD 40 in both the groups, between which their expressions were insignificaly different from each other ( P > 0.05) . Conclusion The effect of transinfect-ed DC by rAAV - Her 2/neu on stimulating allogenetic T lymphocyte proliferation in the mixed lymphocyte reaction is better than that of non -transinfacted DC.
