摘 要:目的:探讨MMP-9及TIMP-1在宫颈癌中的表达及生物学意义。方法:建立组织芯片(TissueMicroarry,TMA)技术平台,应用该技术结合免疫组织化学S-P法检测3种蛋白在宫颈癌中的表达。结果:组织芯片制备成功,脱片率为0.025%。MMP-9在正常宫颈(NCE)、宫颈上皮非典型增生(CIN)和宫颈浸润癌(1CC)的阳性率分别为21.8%、50.0%和81.7%,ICC、CIN分别与NCE比较有统计学意义(P<0.05)。TIMP-1在3组中的阳性率分别为21.9%、25.0%和27.96%,3组间无显著性差异(P>0.05)。宫颈癌中MMP-9的表达与病理分级有关(P<0.05)。伴有盆腔淋巴结转移者MMP-9阳性表达率显著增高(P<0.05),而TIMP-1表达阳性率显著降低。宫颈癌中MMP-9表达高于TIMP-1者,其盆腔淋巴结转移的发生率显著高于MMP-9与TIMP-1表达平衡者(P<0.05)。结论:建立了组织芯片,为研究宫颈癌的生物学行为提供了有力的技术平台。宫颈癌MMP-9表达增强而TIMP-1表达降低可能使宫颈癌更易发生侵袭转移。联合检测宫颈癌中MMP-9及TIMP-1的表达对进一步了解宫颈癌生物学行为和判断其预后有一定的价值。Objective:To investigate the expression of MMP-9 and TIMP-1 in cervical carcinoma and its biological significance.Methods:The technique platoform of tissue microarray (TMA) was established.The expression of 3 proteins in cervical carcinoma was detected by TMA and immunohistochemical SP methods.Results:The preparation of TMA was successful.The rate of shedding slice was 0.025%.The positive rate of MMP-9 in normal cervical epithelium (NCE),cervical atypical hyperplasia (CIN) and invasive carcinoma of cervix (ICC) was respectively 21.8%,50.0% and 81.7%,which in ICC and CIN was statistically differed from that in NCE (P<0.05).The positive rate of TIMP-1 in NCE,CIN and ICC was 21.9%,25.0% and 27.96%,respectively.There was no difference among three groups(P>0.05).The expression of MMP-9 in cervical carcinoma was associated with pathological grades (P<0.05).In cases with pelvic lymphonade metastasis,the positive rate of MMP was increased and that of TIMP-1 was decreased.The incidence of pelvic lymphonade metastasis was higher in cervical carcinoma with higher MMP-9 than TIMP-1,compared with that with balanced expression of MMP-9 and TIMP-1(P<0.05).Conclusion:TMA provides effective technique platform for studying biological behaviour in cervical carcinoma.The cervical carcinoma with expression of MMP-9 and lower expression of TIMP-1 is apt to metastasize.The combined detection of MMP-9 with TIMP-9 is valuable in further comprehending the biological behaviour and prognosis of carvical carcinoma.
