摘 要:目的:研究右归丸对大鼠卵巢卵泡细胞凋亡的影响与作用机理。方法:以雌性SD大鼠注射糖皮质激素造成肾阳虚模型,采用免疫组织化学方法观察了右归丸对肾阳虚大鼠卵巢的卵泡细胞TNFα、Caspase3、Bcl2、Bax表达的影响。结果:在对大鼠卵巢卵泡细胞Bcl2表达的影响上,右归丸低剂量组、金匮肾气丸组与模型组存在明显的差异(P<0.05),右归丸低剂量组、金匮肾气丸组可通过上调bcl2水平,发挥抑制凋亡的作用(P<0.05),TNFα、Caspase3、Bax也存在相应改变。结论:提示右归丸温阳补肾、填精补血的现代机理之一,可能与该药调节卵泡细胞凋亡途径上的Bcl2、Caspase3、Bax表达有关。同时中成药金匮肾气丸也具有相似的作用。Objective:The effect of Yougui Bolus(YGB)can tonify kidney-yang and blood.To Study the effects of YGB from the changes of apoptotic Expressions in the ovary cell of kidney-yang deficiency rats,so as to explore the molecular mechanism of YGB in treating irregular menstruation amenrrhea and infertility on.Methods:Female SD rats was taken as the kidney-yang deficiency model with injecting glucocorticoid,the immunohistochemical assay was applied to observe the expression of markers such as TNFα、Caspase-3、Bcl-2、Bax in the ovary cells of the rats.Results:To the effect of the Bcl-2 expressions in ovary cells of the rats,The YGB lower dosage group and Jingui bolus groups have obviously differences from the model groups.(P<005),YGB lower dosage group and Jinguishenqi bolus groups could inhibit the apoptosis by improving Bcl-2 level,in the same time TNFα、Caspase-3、Bax have the corresponding changes.Conclusion:This results indicated that the mechanisms of YGB maybe regulate the expressions of Bcl-2、Caspase-3、Bax on the ovary cell apoptotic road,which possibly is one of mechanisms of YGB in treating delayed menstruation or hypomenorrhea、amenorrhea and infertility.Jinguishenqi bolus groups showed the similar results also.
