摘 要:目的探讨热休克蛋白72(heat shock protein,HSP 72)对大鼠急性高眼压性视网膜神经节细胞(RGCs)损伤的保护作用。方法右眼前房内灌注平衡盐溶液制作大鼠急性高眼压性 RGC 损伤模型;用热休克反应或腹腔内注射硫酸锌诱导模型大鼠 RGCs 中 HSP 72表达,观察 HSP72表达以后,模型大鼠 RGC 密度的改变情况,并与未处理的模型大鼠 RGC 密度相比较。结果正常大鼠 RGC 密度为2 504±181个/mm^2;急性高眼压模型大鼠 RGC 密度为2 015±111个/mm^2。热休克反应组和硫酸锌注射组 RGC 密度分别为2207±200个/nm^2和2262±155个/mm^2,显著高于模型组(P_(热体克)=0.036,P_(硫酸锌)=0.019);HSP 72阴性对照组与模型组 RGC密度差异无显著意义(P_(槲皮黄酮+热休克)=0.260,P_(槲皮黄酮+硫酸锌)=0.748)。结论热休克反应和腹腔内硫酸锌注射在急性高眼压模型大鼠 RGCs 中诱导产生的 HSP 72;可能对急性高眼压性 RGC 损伤具有保护作用。Purpose To investigate whether the induction of HSP 72 by heat stress(HS)or zinc sulfate(Zn^(2+)) administration can increase survival of RGCs in rat model of acute glaucoma.Methods Acute glaucoma models were made by intraeameral irrigation with BSS at 102 mmHg tor two hours in right eyes of male Wistar rats.Model rats were treated by heat stress once a week or intraperitoneal injection of zinc sulfate(24.6 mg/kg)every two weeks,two days after intracameral irrigation.In cuntrol groups,quercetin(400 mg/kg)was intraperitoneally injected to inhibit the induction of heat shock proteins 4-6 hours before heat strss or zinc administratian.Treated model rats were sacrificed two weeks later after treatment, untreated rats were sacrificed 16 days following intracameral irrigation.Nissl staining was performed with 1% toluidine blue, RGCs were counted under microscope.Results The average RGC denstiy was 2 504±181 cells/nun^2 in normal Wistar rats and 2 015±111 cells/mm^2 in glaucoma rats.The average RGC densities were 2 207±200 cells/mm^2 in heat group,2 262± 155 cells/mm^2 in zinc sultate(Zn^(2+))group,there were significant differences in RGC densities amnng all groups(P= 0.040,kruskal-Wallis test).HS and Zn^(2+)group had statistically great RGC densities than model group(P_(HSR vs.model)= 0.036,P_(Zn^(2+)_(vs.model)=0.019,Mann-Whitney test).However,no significant difference in RGC densities was found between HSP 72 negative control group and model group(P_(HSR+que vs.model)=0.260,P_(Zn)^(2+)+(que vs.model)=0.748;Mann-Whitney test). Conclusion The results demonstrated that the induction of HSP 72 in RGCs by heat stress or zinc administration plays an important role in RGC survival in rat acute glaucoma.
