摘 要:目的:研究口腔鳞癌组织中促血管生成素2(angiopoietin2,Ang2)的表达及其与微血管密度(microvesseldensity,MVD)的关系,探讨Ang2在口腔鳞癌血管生成中的作用及意义。方法:用免疫组织化SABC法检测41例口腔鳞癌标本(有淋巴结转移者13例)及10例正常口腔黏膜组织中的Ang2和CD34表达并计数微血管密度。结果:Ang2在口腔鳞癌组织中阳性表达率为68.29%(28/41),显著高于Ang2在正常组织中的表达(P<0.05);Ang2表达主要集中于癌灶边缘的血管重建区。口腔鳞癌组织中MVD显著高于正常组织(P<0.01);中分化型的肿瘤MVD值显著高于高分化型的肿瘤(P<0.05);有淋巴结转移的肿瘤MVD值显著高于无淋巴结转移的肿瘤(P<0.05)。Ang2表达阳性组中MVD明显高于Ang2表达阴性组(P<0.01),Ang2的表达与MVD呈正相关(P<0.01)。结论:Ang2在口腔鳞癌组织中的高表达可能在口腔鳞癌的发生发展中起重要作用,并与肿瘤血管生成有关。Objective:To study the role of angiopoietin-2(Ang-2) in angiogenesis in oral squamous cell cacinoma(OSCC). Methods:Expression of angiopoietin-2 and CD_34 in 41 cases of OSCC(13 cases with lymph node metastasis)and 10 cases of normal oral mucosa was detected by immunohistochemical SABC method. Microvessel density(MVD) in the samples was counted under microscope. Results:Ang-2 was detected in 28 out of 41(68.29%) cases of OSCC and 2 out of 10(20%) of normal oral mucosa(P<0.05). The MVD was 43.66±8.92 in OSCC and 26.60±5.99 in normal oral tissues (P<0.01). The MVD was 47.13±8.21 in moderately differentiated tumor and 41.44±8.79 in highly differentiated tumor (P<0.05).That in tumor with lymph node metastasis and in tumor without lymph node metastasis was 47.77±7.00 and 41.75±9.17 respectively(P<0.05). The MVD was 46.07±8.38 in Ang-2-positive OSCC and 38.46±8.02 in Ang-2-negative OSCC (P<0.01).MVD was positively correlated with the expression degree of Ang-2(P<0.01). Conclution:Overexpression of Ang-2 in OSCC is closely associated with the tumor angiogenesis and plays a potential role in the development and progression of OSCC.
