离体热暴露大鼠心肌细胞内钙及Ca^(2+)-ATP酶活力的变化

摘  要:观察了离体成年大鼠心室肌肌质网、线粒体Ca2+-ATP酶活性和钙含量及培养乳鼠心肌细胞内游离钙浓度(心肌[Ca2+]i)在不同温度热暴露40min后的变化。结果表明:热暴露后,心肌匀浆、肌质网及线粒体中的Ca2+-ATP酶活力有所下降,心肌肌质网及线粒体中的钙含量亦有降低趋势,心肌[Ca2+]i显著升高。提示:热暴露后心肌细胞钙超载是导致机体心功能损害的重要原因之一。The activity of C2+-ATPase,calcium content of sarcoplasmic reticulum SR and mitochondria of myocardium in adult rats and intracellular Ca2+in primary culture cardiomyocytes prepared from neonatal rats were observed after exposure to 37 ℃, 39℃, 41℃,and 43℃ for 40 min respectively in vitro.The activity of Ca2+-ATPase was measured by hydrolysis of ATP to release inorgnic phosphorus,the intracellular Ca2+by the fluorescent Ca2+indicator,Fura-2/AM.The results showed that the calcium content and activity of Ca2+-ATPase in SR and mitochondria decreased gradually as exposure tempeature increased, the activity of Ca2+-AT-Pase in SR and mitochondria decreased by 28.8%, 26.9%,respectively at 43℃.The intracellular Ca2+concentration increased by 16.7%,26.5%,and 46.0%respectively at 39℃,41℃,and 43℃ It is suggested that the decrease of activity in Ca2+-ATPase may lead to the rise of intracellular Ca2+which maybe one of the cause of cardiac function disorder or heart failure in heat stress.
