人乳腺癌c－erbB2基因扩增及过度表达的研究

摘　　要：应用定量ＤＮＡ点杂交法及免疫组化法对１０１例石蜡包埋乳腺癌组织ｃ－ｅｒｂＢ２原癌基因的扩增及过度表达进行了研究，结果显示，３４．７％的乳腺癌病例有ｃ－ｅｒｂＢ２基因扩增，２３．８％的病例有该基因编码蛋白的过度表达。基因扩增及蛋白表达虽有相关关系，但并不完全相吻合。较高水平的ｃ－ｅｒｂＢ２蛋白过度表达与肿瘤大小及淋巴结转移数呈显著正相关。原发部位及淋巴结转移乳腺癌中，该基因及表达的改变无明显差异，说明ｃ－ｅｒｂＢ２基因型及表型的改变出现在肿瘤转移之前，而在转移过程中保留。AbstractQuantitative DNA dot blot hybridization and immunohistochemical methed were employed to de-tect amplification and overexpression of proto-oncogene c-erbB2 in 101 paraffin-embedded breast can-cers.The results showed that c-erbB2 was amplified and overexpressed in 34.7%and 23.8%of ex-amined cases respectively.The overexpression of c-erbB2 was correlated with,but not always paralleto the gene amplification.The moderate to high-level overexpression of c-erbB2 detected by immuno-histochemistry was significantly associated with the size of the tamour and the nuber of lymph nedesinvolved.No differences were found in the alteratio of c-erbB_2 gene and its protein expression betweenprimary and metastatic breast cancers indicating that the genotypic and phenotypic changes of c-erbB2occured prior to and maintained in the process of metastasis of breast cancer.
