摘 要:我们前已报道用抗HBVx抗原(HBxAg)三种不同肽段的抗体检出肝癌细胞中HBxAg的存在。我们用PCR技术扩增肝癌肝组织中的x基因,用重组HBxAg及其抗体分别检测肝癌患者血清中的抗HBx和HBxAg并用免疫组织化学的方法分析肝组织中HBxAg的表达及其分布。我们发现HBxAg在肝癌肝组织中阳性率高达92.6%而且分布广泛,主要表达在细胞浆,但细胞核内亦有见到。癌旁组织检出率也很高,而且在小胆管上皮细胞内亦见有HBxAg的存在。In our previous papers intrahepatic localization of HB xAg in liver cancer was investigated usingantibodies against peptide fragments representing three particular regions of X gene sequence. In thisstudy PCR technique was used to detect the X gene in the liver tissue sections.The antisera against therecombinant HBxAg which was synthesized in E. coli were prepared andused for the detection ofHBxAg in the liver cell of 7 8 patients with HCC. And the HBxAg/anti HBx were also analizedin the sera from the patients using ELISA. It was found that HBxAg was not only seen in hepatocyte cyto-plasm but also in nuclei and the total positivity rate of HBxAg is 92.6%(75/78). Besides this, we alsofound the predominant expression of HBXAg in bile duct ethelial cells. Our findings show that theexpression of HBxAg closely related to the development of HCC.
