摘 要:对小鼠肺腺癌细胞系(LA-795)高、低转移亚系(LA-795-C_6和LA-795-C_7)体外培养第20及30代细胞染色体G显带核型进行比较,结果高转移亚系细胞染色体众数较低转移亚系细胞具有不稳定性的特点,表明转移潜能的差异和遗传物质的稳定程度密切相关.高、低转移亚系出现相同及不同的标志染色体和异常染色体,其中,高转移亚系出现标志染色体8种及del(8)q ̄(ter),del(12)q ̄(ter),11P ̄+,13P ̄+,tri(2M4),dic(2M5),M6:7q ̄+?;而低转移潜能亚系出现标志染色体12种及del(8)q ̄(ter),del(12)q ̄(ter),r(15),dic(M6;2M7),M8:7q ̄_?t(2:2),t(2:15),t(15:M10),t(15:M11),t(5:M12).表明来源于同一母系而转移潜能不同的亚系染色体核型具有本质上的差异,转移潜能与其密切相关.另外,分析低转移亚系出现t(2:2);t(C2:15)t(15:M10);t(15:M11); t(5:M12);r(15)等现象,推测高、低转移潜能亚系细胞可能存在本身H-2系统及CD44抗元表达的差异,这很可能?LA-795-C_6 and LA-795-C_7,the low and high metastatic sublines isolated frommouse lung adenocarcinOma were studled comparatively with G-banding technlque atpassage 2Oth.and 3Oth.The model dllDmOsoase number of high metastatic subline(LA-795-C_6was more ul1stable than that of fow imtastatic subline(LA-795-C_7)This asult showed thatthe degree of the instability of cytogenetic subetance was closely related to metastatic potentialJtis visible that the same and different marker chromosomes as well as the same and differntchromosomes aberration preexisted in the fow and ingh metastatic sublines,In the highmetastatic subline,there were eight types of marker chromosome,and the chromosomeabecation was del(8)q ̄(ter),del (12)q ̄(ter),11P ̄+, 13P ̄+tri(2M5)and M6:7q ̄+? However,in thelow metastatic subline,there were twelve types of marker chromosome,and the chromosomeabenstion was del(8)q ̄(ter)、del(12)q ̄(ter),3P ̄+,r(15),dic(M6;2M7),M8:7q ̄+?t(2:2),t(2:15),t(15:M 10),t(15:M 11),t(5:M12).All these results demonstrated that the karyotypes of the different metastaticsubpopulations isolated from the parent cell line(LA795)were different, and the heterogeneouskaryotype was refated to the metastatic potential.In addition,the phenomenon that lowmetastatic subline showed both chromosome No.2 in translocation with chromosomes Nos.2and 15 sepately,both chromosomes Nos.15 in translocation with marker chromosome Nos.10 and 11 separately,and chromosome No. 5 inthenslocation with marker chromosome No.12.Itwas deduced that the expression of H-2 gene prodgcls prodgcls and CD44 in the low and highmetastatic sublines (LA-795-C_6/LA-796-C_7)might be different,which were correlated with it' s metastatic property. From all these resullts we concluded in brief:to study deeply andintensively on the marker chromosomes and chromosomes aberration discovered in the experiment.One might find a key to the mechanism of metastasis.
