摘  要:应用PCR技术检测细小病毒H-1DNA在人肝癌与裸鼠正常组织中复制的差异黄青山,马承武,郭兰萍,陈献华,罗祖玉(上海复旦大学生理与生物物理学系,上海200433)关键词:自主性细小病毒H-1及MVM,聚合酶链式反应(PCR),人肝癌模型,抑瘤作用,肿...highly sensitive and specific PCR method for the detection of the autonomousParvoviruses H-1 (H-1) and minute virus of mice (MVM) was established by the constructionof two primers, P3 and P4, copying sequences encoding the non-structural (NS) proteins ofH-1 and MVM. The method had been used successfully for monitoring H-1 replicationboth in in vitro system, NB-K cells and human hepatonia cells, QGY-7703, and in thetu-mor-bearing nude mice. Several pfu of H-1 could be detected in vitro by a specific ampli-fied H-1 DNA band (908bp), and 6 hrs after H-1 infection of NB-K cells. The virus spe-cific DNA band was detected in DNA extracted from ca 250 infected cell. H-1 (1×10 ̄8pfu)inoculation of the tumor-bearing nude miee was accmplished by intravenous injectionthrough the caudal vein, and DNAs were extrdcted from the human hepatoma or the normaltissues of nude mice at various time intervals after inoculation. The results showed that H-1replication was found exclusively within the confines of the transplanted human hepatoma butnot in any of the normal tissues of the nude mice.
