摘  要:用WQ-9E钾离子透入仪筛选出对痛刺激敏感的SD大鼠8只,随机分成对照组及针刺组,每组4只。针刺鼠用WQ-9E电针仪刺激两侧"足三里"穴,持续30min。所有动物同时用1%戊巴比妥钠腹腔内麻醉后开胸,用Bouin液经心灌注固定,开颅取脑,作连续冰冻切片(片厚40μm),隔片取一。所有切片均按Sternberger(PAP)法进行免疫细胞化学反应,DAB呈色,封片后直接连IBM286-CAP530图像分析系统对视上核,室旁核催产素免疫反应阳性物质作定量测定。结果显示:针刺鼠下丘脑视上核,室旁核催产素免疫反应阳性物质的总面积比对照鼠分别增加了44.1%、47.3%(P<0.01)。提示,催产素免疫反应阳性物质的增加与电针刺激足三里穴有关,催产素可能参与电针镇痛的中枢调节过程。Eight adult male Sprague-Dawley rats sensitive to pain induced by potassium ionophoresis were randomly divided into experimental group and control group. There were four rats in each group. The experimental rats were electroacupunctured 'Zusanli' for 30min. The control rats remained unstimulated. All the experimental and control rats were then aneshetized with sodium pentobarbital, perfused transcardially with Bouis's fixative solution. Then the brains were removed and cut coronally with a freezing rotary microtome (40 μm thick). All the sections of two groups were incubated in anti oxytocin serum and processed for immunostaining by applying the peroxidase-antiperoxidase (PAP) method of Sternberger's.The total areas of oxytocin-immunoreactive (OT-ir) substance of the supraoptic nucleus (SON) and paraventricular nucleus (PVN) on the same side of all sections were measured by a microcomputer image analysis system (IBM286-CAP530). The result showed that the total areas of OT-ir substance of SON and PVN in the experimental rats were significantly increased by 44.1% and 47.3% as compared with the control group respectively(P<0.01).Our data suggest that OT is possibly involved in the central modulation of acupuncture analgesia.
