摘 要:作者用白蛋白包埋法制成具有磁响应的秋水仙碱-白蛋白-磁微粒,并采用MTT法检测了秋水仙碱-白蛋白-磁微粒及游离秋水仙碱对EMT_6细胞的生长抑制作用,对乳癌EMT_6细胞裸鼠移植瘤进行基底部注射和外加磁场定位治疗.结果表明:秋水仙碱-白蛋白-磁微粒及游离秋水仙碱在体内、体外实验均对肿瘤细胞有明显的杀伤作用,体外实验在100μg/ml剂量时,灭活率分别为56.16%和58.46%.对乳癌EMT_6细胞裸鼠移植瘤的抑瘤率分别为32.8%和23.6%,提示秋水仙碱-磁微粒外加磁场引导定位对裸鼠乳腺癌有较好的抑制作用.In the present study albumin-embedded colchicine albumin magnetic microsphere(Fm-HSA-Co)was prepared. MTT was employed to determine Fm-HSA-Co and free colchicine’s inhibition of EMT_6 cellgrowth.Basilar injection and external magnetic orientation therapy were performed on mammary cancerEMT-6 cell xenografts in nude mice.The results showed:Fm-HSA-Co and free colchicine caused evident de-struction of tumor cells both in vitro and in vivo.In the in vitro experiment,the inhibition rates were respec-tively 56.16%and 58.46%(P<0. 01 vs control)at the dose of 100 μg/ml, while in vivo the tumor-inhibit-ing rates against mammary cancer EMT_6 cell xenografts in nude mice were 32.8%and 23.6%(P<0.05 vscontrol),suggesting that with the assistance of external magnetic orientation colchicine albumin magnetic mi-crosphere has stronger inhibition to breast tumor in nude mice.
