K_(562)细胞及K_(562)/ADM耐药细胞内cAMP及cGMP含量的测定

摘  要:测定正常人淋巴细胞内环磷腺苷(cAMP)、环磷鸟苷(cGMP)含量和人红白血病细胞株(K562)及其耐阿霉素细胞株(K562/ADM)的增殖状况和细胞内cAMP、cGMP含量。结果K562与K562/ADM细胞内cAMP含量均低于正常人淋巴细胞内cAMP,但无显著差异,K562与K562/ADM之间细胞内cAMP的含量有显著差异,cGMP含量均未见差异。K562/ADM细胞的增殖状况与cAMP含量呈负相关(r=-0.5823,P<0.05),加入逆转耐药的药物潘生丁、维拉帕米后,其增殖速度下降且差异显著(P<0.05),cAMP含量呈上升趋势;K562细胞也有此改变,但两种药物对两种细胞的影响程度不同。The levels of cAMP and cGMP in normal lymphocyte,K562 and K562/ADM cells were measured,and the rates of growth of K562 and K562/ADM were determined also.The results showed that,the levels of cGMP in three kinds of cell were not different,while the levels of cAMP in K562 and K562/ADM were lower then that in normal lymphocyte,but the difference was not significant.The difference of the level of cAMP in K562 and in K562/ADM was significant.For K562/ADM,negative correlation existed between the rate of growth and the level of cAMP,The growth rate decreased noticeably and cAMP level tended to increase after dipyridamole or verapamil were added.Same changs took place for K562 cell.However,the effects of two drugs on K562 and K562/ADM were different.
