摘 要:以甘蓝型油菜品种(系)和杂种F1代为材料,研究了供试材料和培养基成分对小孢子胚产量(以每花药产生的胚状体个数表示)的影响。不同材料胚产量有显著差异,变化范围为0~12.86。通常,杂种F1代胚产量较品种(系)高。同一材料不同单株间胚产量亦有差异。培养基成分中,大量元素以NLN为最佳,1/2MS和1/2B5培养基上亦可形成少量胚状体;培养基的pH以5.8为佳,提高pH至6.8和7.8,显著不利于胚状体形成和发育;不同材料要求的蔗糖浓度、激素种类和浓度也不同。Varieties and hybrids F1 of Brassica napus were used as materials and their microspores were isolated and cultured.It has been shown that the embryoid yield(No.of embryoid per anther inoculated)vaired with the different materials used,in a range of 0.00~12.86.As a rule,all the hybrids had a higher embryoid yield than the varieties tested.Significant differences in embryoid yield among plants of the same donor material were observed.The basic element of NLN with pH 5.8 was best for embryoid formation among the media tested.It was harmful for embryoid development to increase pH to 6.8~7.8.The optimun sucrose concentration and types of hormone in the media varied with different materials.
