摘  要:本文观察了血小板源生长因子(PDGF)A、B链及其α、β受体基因表达在低氧大鼠肺中的变化。Northern印迹杂交显示正常大鼠肺中可表达PDGF-A链、B链和PDGF-α受体、β受体的mRNA。随着低氧时间的延长,mRNA水平迅速增加。PDGF-A、B链mRNA均在低氧的第二天达高峰,分别为正常的1.51倍及1.69倍(均为P<0.05),且B链mRNA基本维持在该水平至第七天。PDGF-α、β受体的mRNA在低氧第四天时达高峰,分别为正常的1.70倍及1.34倍(均为P<0.05)。提示PDGF受体及配体基因的依序和/或合并表达增强在低氧性肺血管结构重建中起一定作用。his study examined the changes of platelet-derived Growth Factor(PDGF)receptor and itsligand genes expression in hypoxic rats lungs. Northern blots analysis revealed that normal lungexpresses PDGF-A,-B chain and PDGF-α,-β receptor mRNA. With the longer of hypoxic,thelevels of mRNA increased rapidly, The mRNA of PDGF-A,and B chain reached a maximum atday 2,respectively 1. 51 fold and 1.69 fold as compared with control,(P<0.05 ),and B chainsustained at this high level to day 7.The mRNA of PDGF-α and-β receptor reached a maximumt day 4,respectively 1.70 fold and 1.34 fold as compared with control(P<0.05). Results suggested that increases of PDGF receptor and its ligand genes expression may becoordinately and subsequently involved in hypoxic pulmonary vascular remodeling.
