摘  要:目的探讨表达谱芯片技术在罗勒多糖抗肿瘤机制研究中的价值与作用。方法建立荷NuTu-19卵巢癌大鼠模型共20只,10只予罗勒多糖(2.5mg/kg)连续灌胃50d(药物组),另10只予等量生理盐水灌胃50d(对照组)。以腹水量、腹腔脏器转移程度积分判断各组肿瘤生长及转移情况。分别用Cy3-dUTP、Cy5-dUTP标记两组癌组织cDNA,与含有2000点的大鼠基因表达谱芯片杂交。结果对照组与药物组共有168条基因出现差异表达。结论表达谱芯片技术可以快速确定药物作用的可能机制,并为进一步研究提供重要信息。Objective: To explore the application of cDNA microarray in the mechanism analysis of drug.Methods: 20 rats bearing NuTu-19 cells were randomly divded into two groups,the drug group (n= 10) were treated with basil polysaccharide for 50d,while the control group (n= 10) treated with physiological saline at same dose,changes of the biology phenotype of tumor cell were observed. Total RNA of the tumor tissue was extracted ,and the fluorescent cDNA probes were prepared through reverse transcription of the isolated mRNAs.The RNA samples were labeled with Cy3-dUTP or CyS-dUTP, then hybridized with the microarrays. Results:Compared with the control group, 168 genes in the drug group showed different expression. Conclusion:cDNA microarray technology may be power tools to discover the possible mechanism of drug in gene level,and can provide new informations to accelerate the development of new drugs.
