摘  要:目的采用体外细胞培养,免疫组织化学染色和放射免疫测定方法,研究α干扰素(αIFN)对体外培养人垂体功能性腺瘤细胞激素分泌的影响。方法对11例垂体生长激素(GH)及GH/催乳素(PRL)腺瘤和15例垂体PRL腺瘤进行体外细胞培养,观察细胞形态的改变和生长特点。采用免疫组织化学染色技术研究αIFN和αRIFN在侵袭性和非侵袭性垂体腺瘤、功能性和非功能性垂体腺瘤之间阳性表达率差异。结果大多数垂体腺瘤细胞有αIFN和αRIFN表达,尤其在侵袭性垂体腺瘤细胞中呈高表达;αIFN对垂体腺瘤细胞激素分泌有较强的抑制作用;αIFN对大多数溴隐亭(BC)或奥曲肽(SMS)耐药的腺瘤细胞仍有抑制激素分泌效应;αIFN能增强BC对垂体PRL腺瘤分泌的抑制作用。结论αIFN具有用于临床治疗功能性垂体腺瘤的可能性。Objective To study the effects of α-IFN, BC and SMS on hormone secretion of the invitro cultured cells of functioning pituitary adenomas and explore the possihilities of biological therapy of pituitary adenomas. Methods In in vitro cell culture, immunohistochemistry staining and radioimmunoassay were used to observe the proliferation of the cultured cells, detect the expression of α- IFN and αR-IFN, and measure the hormone secretion respectively in 26 cases of pituitary adenomas. Results Most of pituitary adenoma cells expressed α-IFN and αR-IFN, especially in invasive pituitary adenomas.The secretion of GH and PRL by pituitary adenoma cells could be inhibited by α-IFN. Hormone secretion in the majority of BC-or SMS-resistant pituitary adenoma cells could be significantly inhibited by α-IFN.α- IFN could enhance the inhibitory effects of BC or SMS on hormone secretion. Conclusion It is possible that α-IFN can be used to treat functioning pituitary adenomas in clinical practice.
