摘 要:背景和目的:多聚免疫球蛋白受体(polymericimmunoglobulinreceptor,pIgR)基因在机体免疫应答过程中发挥重要作用,并且与EB病毒(Epstein鄄Barrvirus,EBV)的感染途径密切相关,因而是研究鼻咽癌(nasopharyngealcarcinoma,NPC)遗传易感性的理想候选基因。本研究旨在探讨pIgR基因与NPC遗传易感性之间的关系。方法:针对预选的4个单核苷酸多态性位点(singlenucleotidepolymorphisms,SNPs),通过PCR产物直接测序,对广东地区的528例NPC患者及408例健康人血标本DNA进行基因分型,然后采用病例鄄对照的研究方法分析等位基因频率之间的差异。结果:4个SNPs总体的基因频率分布在病例鄄对照之间无显著性差异(P>0.05);按45岁年龄分组分析,在小于等于45岁年龄组,多态性位点C8880T(Gly199同义突变)的次要等位基因T的频率在NPC患者中为7%,在健康人中为4%,病例与对照之间具有显著性差异(P<0.05),OR值(1.84)也提示携带次要等位基因T的个体患病风险增加。结论:pIgR基因外显子区域多态性位点C8880T与NPC的易感性相关,pIgR可能在NPC的发生发展中起一定作用。BACKGROUND & OBJECTIVE: Polymeric immunoglobulin receptor (plgR) gene plays a central role in immune response, and is closely related to Epstein-Barr virus (EBV) infection. Therefore, it is an ideal candidate gene for research on genetic susceptibility of nasopharyngeal carcinoma (NPC). This study was designed to explore correlation of plgR gene polymorphisms to genetic susceptibility of NPC. METHODS, Four single nucleotide polymorphisms (SNPs) within the exon regions of plgR were detected by polymerase chain reaction (PCR); plgR was genotyped in 528 NPC patients and 408 healthy individuals in the Cantonese population to analyze frequencies of its allelic genes. RESULTS: Differences in frequencies of the 4 SNPs between NPC patients and healthy controls were not significant(P〉 0.05). All subjects were categorized into 2 groups by age of 45;of the subjects in age of ≤45 group, the frequency of minor allele T of SNP C8880T was significantly higher in NPC patients than in healthy controls (7% vs. 4%,P〈 0.05). Individuals carried allele T suffered higher risk of NPC (odds ratio =1.84). CONCLUSION-SNP C8880T of plgR is related with NPC susceptibility; plgR gene may be associated to risk of NPC development.
