摘 要:目的 探讨肝母细胞瘤组织Fas、FasL及FAP-1和组织中淋巴细胞的Fas、FasL的表达状况及临床意义。方法 应用Fas、FasL多克隆抗体及FAP-1单克隆抗体对15例小儿肝母细胞瘤组织标本进行免疫组化染色,以检测其组织Fas、FasL及FAP-1和淋巴细胞的Fas、FasL的蛋白表达,并以相应的正常肝组织作对照。结果 肝母细胞瘤组织中Fas表达(78%)高于正常肝组织(5%),阳性信号主要位于细胞膜和细胞浆,呈胞浆型表达。正常肝组织中FasL表达阴性,而肝母细胞瘤中FasL表达73%,两者表达率差异明显;在15例肝母细胞瘤中FAP-1阳性表达13例。同时发现瘤周淋巴细胞Fas、FasL的阳性表达率分别为80%、68%,而在正常肝组织中的表达均为阴性。结论 正常肝组织同时表达Fas/FasL,在维持其自身细胞的增殖与凋亡的平衡中起重要作用;肝母细胞瘤组织Fas、FasL表达上调以及FAP-1阳性表达可诱导表达Fas的淋巴细胞凋亡以实现免疫逃逸。AIM To investigate the expression of Fas,FasL and FAP- 1 protein in hepatoblastoma and the xpression of Fas,FasL in lymphocyte and its clinical significance. METHODS The expression of Fas,FasL and FAP- 1 was examined in 15 cases of hepatoblastoma and in 15cases corresponding normal liver tissue with Fas,FasL and FAP - 1 clonal antibody respectively ,by immunohistochemical technique. RESULTS Fas expression rate was higher in hepatoblastoma(78% )than that in normal liver tissue(5% ), the staining signal of Fas was mainly located in cytoplasm and membrance. FasL expression in normal liver tissue was negative, but, it is 73 % in hepatoblastoma, and the imparity of expression rate between them is very obvious. That the expression of FAP - 1 was found in all hepatoblastoma is 15 cases. And in this test, we all sofound theexpression rate of Fas,FasLof lymphocyte 80% ,68%, they are negative in normalliver tissue. CONCLUSIONS Coexpression of Fas/FasL play an important role in the balance of proliferation and death in normal liver tissue. The expression of Fas and FasL rise and the expression of FAP- 1 in hepatoblastomacolorectal cancer escapes immune survillance.
