血糖对缺氧缺血新生大鼠脑Bcl-2表达的影响

摘  要:目的应用竞争性RTPCR技术,研究不同血糖浓度对缺氧缺血新生大鼠脑内的细胞凋亡抑制基因Bcl2基因表达的影响。方法将7日龄SD新生大鼠按发生缺氧缺血后的时间进行分组。结果缺氧缺血前重度高血糖组的基因表达时间较缺氧缺血组至少延迟12小时,且表达强度更甚于缺氧缺血组。缺氧缺血前低血糖组的基因表达则显著弱于缺氧缺血组。结论提示缺氧缺血前低血糖对缺氧缺血新生大鼠的脑损害作用,部分可能是通过对Bcl2基因的负向调节而实现,而缺氧缺血前重度高血糖对缺氧缺血新生大鼠的脑保护作用,部分可能是通过对Bcl2基因的正向调节而实现。Objective To investigate effect of different concentrations of blood glucose on expression of Bcl-2 gene, using competitive RT-PCR.Methods The newborn SD rats aged 7days was divided into eight groups according to times when HI occured. After establishment of eight HI models (including normal 0ontrol group and false operation group), the rats were killed and their brain tissues sample were taken. Changes in expression of Bcl-2 in brain tissues in each model group were observed and compared to evaluate objectively cerebral protection of glucose for rate with HI. Results The results showed that expression time of bel-2 gene delayed at least for 12 hours, and in the group with higher hyperglycemia before HI, expression intensity of the gene was stronger than that in HI group. However, expression of the gene in the group with hytpoglycemia before HI was significantly weaker than that in HI group. Conclusion It was suggested that cerebral damage effect of hytpoglycernia before HI canbe probably induced through downregulation of the gene partially, and cerebral protective effect of hyperglycemia before HI can be probably induced through upregulation of the gene partially.
