摘  要:目的:探讨八肽生长抑素奥曲肽(OCT)对三苯氧胺(TAM)所致实验性大鼠性激素内环境失衡的逆转作用。方法:Wistar雌鼠接受致癌剂二甲基苯蒽8周后随机分为对照组和治疗组,治疗组采用OCT,TAM或二者联合治疗14周。应用酶免疫分析法测定血清中雌二醇(E2)、孕酮(P)、泌乳素(PRL)、卵泡刺激素(FSH)和黄体生成素(LH)的水平。结果:血清E2和P含量在TAM组均明显高于对照组(P<0.05),而OCT+TAM组与对照组无明显差异(P>0.05);PRL含量在OCT组、TAM组和OCT+TAM组(1.40ng/ml)均显著低于对照组(2.85ng/ml)(P<0.05,P<0.01)。结论:OCT在体内可以拮抗TAM对内源性雌激素的影响,从而减少因性激素内环境不平衡所致的副作用。Objective:To invcstigate the effect of somatostatin (SST) analog octreotide (OCT) in antagonizing the maladjustment of sexual hormones homeostasis induced by tamoxifen(TAM) in rat. Metkods:Eight weeks after DMBA(a single dose 100mg/kg body weight by subcutaneous injection) application, the female Wistar rats were randomized to control and treatment groups. Rats in treatment groups underwent TAM( 1 mg/kg 5 times weekly for 14 weeks by subcutaneous injection), OCT( 0.1 mg/kg bid for 14 weeks by subcutaneous injection), or the eombinatkm of both. The levels of sexual hormones (estradiol, progesterone, prolactin, follicle stimulating hormone, luteinizing hormone) were assessed by enzyme immunoassay(EIA). Re.suits:The levels of estradiol and progesterone in serum were remarkably higher in TAM group(21.69pg/ml,6.14ng/ml) than in control group(16.92pg/ml,4.13ng/ml) respectively ( P 〈 0.05, P 〈 0.01 ), and there was no significant difference between OCT + TAM group ( 14.58pg/ml, 3.88ng/ml) and control group. The prolactin concentrations in ,serum in the three treatment groups were lower than in control group( P 〈 0.05, P 〈 0.01 ).Conclusion :OCT can antagonize the effect of TAM to ascend the level of endogenous estrogen, and abolish the ill-effects resulting from maladjustment of sexual homones homeostasis.
