流式细胞术PI、Annexin V/PI和APO2.7法检测细胞凋亡的比较

摘　　要：目的:探讨流式细胞术PI法、APO2.7法以及AnnexinV/PI法三种凋亡检测方法的应用价值。方法:使用PI法、APO2.7法以及AnnexinV/PI法三种方法同时对槲皮素处理后的慢性粒细胞白血病K562细胞进行凋亡检测对比研究。结果:三种方法不同时间组间的相关系数分别为0.823(P>0.05),0.949(P<0.05),0.994(P<0.01)。药物作用24小时组APO2.7法与AnnexinV/PI法检测细胞凋亡相关系数为0.925(P<0.05),48小时组相关系数为0.993(P<0.01)。结论:流式细胞术检测凋亡时可以选用AnnexinV/PI法和APO2.7法,不推荐选用PI法。Objective： To evaluate PI, Annexin V/PI and APO2.7 methods for measuring apoptosis by flow eytometry.Methods： The apoptotic percentage of K562 cells treated with quercetin for 24 or 48 hours was measured by flow cytometry using propidium iodide （PI）, APO2.7 and Annexin V/PI double staining methods respectively. Results： The correlation factors between 24 and 48 hours groups of each methods were 0.823 （ P 〉 0.05）, 0.949 （ P 〈 0.05）, 0.994 （ P 〈 0.01 ） respectively.For 24 or 48 hours group, the correlation factor between Annexin V/PI and APO2.7 Methods was 0.925 （ P 〈 0.05） or 0.993 （ P 〈 0.01） respectively. Conclution： Both of Annexin V/PI and APO2.7 methods can be used to measure apoptosis by flow cytometry presently. PI method is not suitable for apoptosis detection.
