摘 要:目的:探讨黄芪对腹膜透析相关性腹膜纤维化的作用。方法:人腹膜间皮细胞(HPMC)做体外培养,第三代细胞用于实验。分5组观察,对照组为RP-MI1640;PDS(peritoneal dialysis solution,腹膜透析液)组为加入PDS;黄芪12、3、组为加入含黄芪注射液的PDS,黄芪终浓度分别为102、04、0 mg.ml-1。分别在24 h采用ELISA法检测细胞上清液中转移生长因子-1β(TGF-1β)的含量,采用逆转录多聚酶链式反应(RT-PCR)检测TGF-1βmRNA表达。结果:腹膜间皮细胞在PDS干预下分泌TGF-1β明显增加(P<0.05)。腹膜间皮细胞经含不同浓度黄芪注射液的PDS干预后,TGF-1β分泌与PDS组比较有显著下降(P<0.05)。不同浓度黄芪注射液组之间比较无显著差异(P>0.05)。腹膜间皮细胞在PDS干预下TGF-1βmRNA表达明显升高,比对照组上升62.43%,腹膜间皮细胞经含不同浓度黄芪注射液的PDS干预后,TGF-1βmRNA表达显著低于PDS组(P<0.01)。结论:商业性腹膜透析液可促进腹膜间皮细胞分泌和表达TGF-1β,黄芪可以部分抵消这一作用所致的腹膜透析相关性腹膜纤维化。AIM: To investigate the effects of Astragalus on peritoneal fibrosis correlating peritoneal dialysis. METHODS: Human peritoneal mesothelial cells (HPMC) were cultured in vitro. They were divided into five groups: RPMI1640 culture medium as control group, PDS( peritoneal dialysis solution) containing 1.5% glucose as PDS group, PDS containing 1.5 % glucose and Astragalus 10 mg·ml^-1 as Astragalus group 1, PDS containing 1.5 % glucose and Astragalus 20 mg· ml^- 1 as Astragalus group 2 and PDS containing 1.5 % glucose and Astragalus 40 mg· ml^-1 as Astragalus group 3 for testing. The levels of TGF-β1 in supematant of the cell culture was detected by the method of ELISA and the mRNA expression of TGF-β1 were tested by the method of reverse transcriptase PCR (RT-PCR) at 24 hours. RESULTS: The level of TGF-β1 was significant lower in control group than that in PDS group (P 〈 0.05). Marked lower level of TGF-β1 was found in Astragalus group 1, Astragalus group 2 and Astragalus group 3 compared with the PDS group ( P 〈0.05). The mRNA expression of TGF-β1 increased significantly to 162.43% compared with the control group. The mRNA expression of TGF-β1 increased 23.52% in Astragalus group 1, 21.36% in Astragalus group 2 and 18.71% in Astragalus group 3, respectively, and significant difference were found when compared with those in PDS groups ( P 〈 0.01 ). CONCLUSION: Astragalus inhibits TGF-β1 protein and mRNA over-expression induced by commercial PDS and plays a beneficial role in peritoneal fibrosis correlating peritoneal dialysis.
