摘　　要：目的探讨羟基喜树碱对人胆管癌QBC939裸鼠移植瘤的基质金属蛋白酶-2(MMP-2)、金属蛋白酶组织抑制剂-2(TIMP-2)的影响,分析MMP-2及TIMP-2与肿瘤发生、发展的关系。方法建立人胆管癌QBC939细胞株裸鼠皮下移植瘤模型,将24只裸鼠随机分成对照组、干预组和实验组,分析移植瘤体生长情况,免疫组化法结合图像分析系统半定量检测瘤体内MMP-2、TIMP-2及MMP-2/TIMP-2的比值。结果与对照组比,实验组可使移植瘤体的MMP-2、TIMP-2含量增高,后者差异有统计学意义(P<0.01);实验组可降低MMP-2/TIMP-2的比值和抑制移植瘤生长,差异有统计学意义(P<0.01)。结论实验组可降低移植瘤MMPV2/TIMP-2比值,对人胆管癌裸鼠移植瘤细胞外基质的降解可能具有抑制作用,进而抑制移植瘤的生长。Aim To investigate the effect of hydroxycamptothecine on matrix metalloproteinase-2 （MMP-2） and tissue inhibitor of metalloproteinase-2 （TIMP-2） in the nude mice by human bile duct carcinoma QBC939 cells in order to analyse the correlation between MMP-2 and TIMP-2 expressions and the development of the tumor. Methods After the model of the transplanted tumor of subcutaneous tissues located the nude mice was established by human bile duct carcinoma QBC939 cells, 24 nude mice were randomly divided into 3 groups, namely control group, interfere group and experiment group. The growth of the transplanted tumor were observed and immunohistochemical staining combined with image analysis were used to determine the expressions of MMP-2 and TIMP-2 in the tumor tissues and the radio of MMPV2/TIMP-2. Results Compared with the control group, the contents of MMP-2 and TIMP-2 in the tumor tissues were increased in experiment group, difference of TIMP-2 （P〈0.05 ） , the ratio of MMP-2/TIMP-2 was decreased and the growth of the transplanted tumor was inhibited in experiment group（ significant difference P〈0.05）. Conclusion Experiment group can reduce the ratio MMP-2/TIMP-2 in the transplanted tumor tissues and inhibited the degradation of the extracellular matrix of the transplanted tumor tissues so as to inhibit the tumor growth.
