摘 要:目的通过调节细胞培养液H+浓度,观察恶性肿瘤细胞外不同H+浓度对TNF-α直接杀伤人肝癌细胞7404作用的影响。方法MTT法于96孔培养板上进行杀伤实验,测定培养液pH值分别为6.6、6.8、7.0、7.2、7.4、7.6状态下,TNF-α对杀伤人肝癌细胞7404的差异。结果pH值为6.6~6.8时,TNF-α对肝癌细胞7404的杀伤率较低,PH值为7.0、7.2、7.4、7.6时,TNF-α对7404细胞的杀伤率逐渐增高,pH值为7.4时TNF-α对7404细胞的杀伤效果最佳。结论TNF-α对肿瘤细胞杀伤作用在弱碱性环境下高于弱酸性环境。Aim By regulating the [H^+] of the cultured cells liquid to observe the effects of different [H^+] on the lethality of TNF-α against human hepatic carcinoma cells 7404. Methods By means of MTT assay ,we will perform killing experiments on 96-hole culture plates. With human hepatic carcinoma cells 7404 as the target cells,the difference of lethality of TNF-α in the case of pH 6.6,6.8,7.0,7.2,7.4 and 7.6 will be assayed. Results Human hepatic carcinoma cells could not grow well when pH is 6.6 or 6.8,but the growth situation is improved when pH rises from 6.8 to 7.4. And in the state of pH6.6 or 6.8 ,TNF-α has the lowest lethality of antitumor . The lethality of TNF-α against the 7404 cells increase when pH value rises,but TNF-α yields the optimal lethality when Ph is 7.4. Conclusion The low-alkali environment provides the TNF-α with optimal state for killing the tumor cells.
