摘  要:为检测人脑胶质瘤浸润淋巴细胞(TIL)的来源及其体外抗肿瘤活性,本实验应用免疫组化方法分别测定10例人脑胶质瘤TIL提取前、提取后及体外经白细胞介素-2(IL-2)刺激增殖后不同时间的细胞成分及其亚型构成。发现TIL主要为T淋巴细胞,其中CD_8阳性细胞占84.29±2.15%,CD_4阳性细胞占8.33±3.43%,B淋巴细胞浸润极少、TIL体外扩增4周.平均增加69±28.17倍,最高达120倍。体外杀伤试验表明,TIL对自体胶质瘤细胞具有高度杀伤力;对异体胶质瘤及K_562细胞也有一定杀伤作用。新鲜分离的TIL无杀瘤活性。经IL-2刺激增殖3周的TIL,杀瘤活性最强。作者认为,从手术切除的脑胶质瘤中提取、扩增的TIL,术后回输入手术瘤床,以杀灭残留的肿瘤细胞,可能成为一种较为理想的过继性免疫治疗方法.In order to investigate the antitumor immune responsiveness of human phenotypic and subtypic characteristies of fresh and recombinant interleukin-2 (IL-2)-expanded TIL were detected by means of the immunohistochemical method. It was discovered that TIL was composed mainly of T lymphocytes showing that CD_8-positive cell was the dominant population being 84. 79±2.15%,CD_4-positive cell was 8. 13±3. 43%,and very few B cells were detected. Cultured for four weeks in vitro, the number of TIL expanded to a mean deviation of 69±28. 17 times, being 120 times higher. Expanded TIL from primary glioma had significantly high cytotoxicity against auto genous tumor cells in vitro, and also had certain cytotoxicity against allogeneic tumor cells and K-562 cells. Freshly isolated TIL had no antitumor effect. The optimal cytotoxicity in vitro was achieved in the third week of culture in the presence of lL-2. All these findings demonstrate that TIL from operative samples of human glioma can be successfully isolated, being cultured in IL-2 and expanded in vitro,The adoptive transfer of TIL into cavity of tumor after operation may be a ideal synthetic treatment of the tumor.
