摘 要:目的 探讨应激引起大鼠脑卒中时细胞与机体防御基因的表达差异以及其在发病学中的意义。方法 制备大鼠高血压脑卒中模型,应用抑制性差减克隆技术对卒中样发作鼠(刺激因素组)和正常对照鼠脑(正常对照组)的差异基因进行筛选。经过两轮杂交后,产生的克隆均为脑卒中鼠脑组织特异表达的序列。每组随机挑取2 88个克隆,进行测序及GenBankBlast生物信息分析。结果 两组共有4 5 6个可用序列,我们对每个与已知基因高度同源的序列以功能为参照进行了分类,发现大鼠卒中后脑组织细胞与机体防御基因表达明显下调(P <0 .0 1) ,2 88个克隆中未发现运输蛋白/膜转运及应激反应相关基因的表达。结论 脑组织运输蛋白/膜转运及应激反应相关基因的表达发生改变,可能是脑卒中发生的分子机制之一。Objective To explore the differential expression of genes for cell and organism defense in brain tissue in stroke-like episodes induced by stress in rats.Methods A hypertension model was established in rats,which produced a complication of stroke. Then, a technique, suppression subtractive hybridization(SSH) was used to identify the differential genes specifically expressed in total rat cerebrum tissue in stroke group and control group. Results Using this SSH technigne, a total of 576 clones were generated from two subtractive libraries, of them 456 were usable and were analyzed. Genes for cell and organism defense in stroke group showed down-regulated ( P 〈 0.01). Genes for carrier proteins/membrane transport and stress response were not detected in stroke group. Conclusions The results suggest that genes for carrier proteins/membrane transport and stress response should induce an increased sensitivity to stroke through the changes of gene expression. Cell and organism defense genes may play important roles in causes and effects of stroke.
