子宫内膜癌异位HCG自分泌观察

摘　　要：[目的]观察子宫内膜癌细胞系异位HCG自分泌现象。[方法]高分化子宫内膜癌EAC和中分化子宫内膜癌JEC两株细胞与乳腺癌细胞株作为观察对象,各用1.5×105个细胞置于1ml不含血清的1640培养液中37℃、5%CO2培养箱内培养,分别于培养后24h、48h和72h提取上清液做HCG放射免疫定量检测。[结果]随着培养时间的延长,两株细胞自分泌的HCG量有所增加,特别JEC株表现更加明显。JEC细胞24h、48h和72h分别为1.83,10.20和28.94,而EAC细胞分别为3.35,4.06和10.34。两株细胞HCG分泌量之间存在线性回归关系(F=4.329,P=0.076)。子宫内膜癌中分化细胞株分泌量高于高分化株。[结论]子宫内膜癌细胞有异位自分泌HCG,分化差的子宫内膜癌的自分泌现象更突出,提示HCG自分泌水平与肿瘤密切相关。[Purpose] To observe the ectopic autocrine phenomenon of HCG in endometrial carcinoma cell lines.[Methods ] Well differentiated endometrial carcinoma cell line EAC and moderately differentiated endometrial carcinoma cell line JEC and breast cancer cell line were investigated. 1.5×10^5 cells of each cell line added to 1 ml culture medium 1640 without serum, and incubated at 37% in a 5% CO2 incubator for 24h, 48h and 72h respectively. The supernatant was collected for HCG assay. [Results] Ectopic HCG increased in EAC and JEC with prolonged culture period, especially in JEC. The quantity of ectopic HCG in JEC after culture for 24h, 48h and 72h were 1.83, 10.20 and 28.94; and in EAC, 3.35, 4.06 and 10.34 respectively. The quantity of HCG in both cell lines is in accordance with linear regression（F=4.329, P=-0.076）. The quantity of HCG secretion in moderately differentiated endometrial carcinoma cell line is higher than that in well differentiated one. [Conclusion] The cell lines of endometrial carcinoma can secrete ectopic HCG, especially in the low differentiation one. It suggests that the level of HCG secretion is closely associated with malignancy.
